Precautions for Tianling ultrasonic cell crusher during use:
1. Ultrasonic parameter setting: set the working parameters of the instrument. For samples with sensitive temperature requirements (such as bacteria), generally use ice bath outside. The actual temperature must be lower than 25 ℃, and protein nucleic acid will not denature.
(1) Time: the ultrasonic time shall not exceed 5 seconds each time, and the gap time shall be greater than or equal to the ultrasonic time, so as to facilitate heat dissipation. The time setting should be based on the principle of short ultrasonic time and more ultrasonic times, which can prolong the service life of the ultrasonic machine and probe.
(2) Ultrasonic power: it should not be too large to avoid sample splashing or foam. If it is less than 10ml sample capacity, the power should be within 200W. Select 2mm ultrasonic probe, and turn the selector switch of the horn behind the panel to the corresponding gear; The power of 10-200ml sample capacity is 200-400W, 6mm ultrasonic probe is selected, and the horn behind the panel is turned to the corresponding gear; For samples with a capacity of more than 200ml and a power of 300-600W, select a 10mm ultrasonic probe, and turn the horn behind the panel to the corresponding gear; (the power of 2mm small probe shall not exceed 350W).
(3) Container selection: select the beaker with the number of samples, which is also conducive to the convection of samples in ultrasound and improve the crushing efficiency. For example; 20ml beaker is used for 20ml processing capacity. For example, 100ml E. coli sample setting parameters: ultrasound for 5 seconds / interval for 5 seconds, 70 times (total time is 10 minutes).
